611 Transcriptomic differences between surgical and non-surgical keloids

Keloids are benign fibroproliferative tumors with features of raised scar tissue and excessive growth beyond the original border. Keloid scars show extracellular matrix(ECM) deposition rich vasculature. Recently, many studies have revealed important pathways keloid pathogenesis. Herein, we focus on difference between traumatic nontraumatic keloids in a transcriptomic level. To preclude intra-lesional heterogeneity, whole was used for bulk RNA-sequencing. Three trauma induced 4 non-trauma related were achieved by surgical excision from 7 different patients. All at stable status without any inflammatory clinical characteristics. RNA extraction sequencing done using an Illumina sequencer. A total 195 differentially expressed genes(DEGs) identified cut-off value p < 0.05, log2FC > 2. Pathway analysis showed that these genes(MMP13, COL5A3) to matrix organization suggesting prolonged ECM formation keloid. For further understanding differences, expanded DEGs based network analysis. With neighbors connected more than 3 very high confidence STRING (confidence score 0.9), (n=461) including neutrophil degranulation, chemokine signaling pathway, JAK-STAT pathway (JAK2, STAT3, p<0.001). Also, HIF-1 genes, which previously noted as hypoxia causing increase collagen, found (p<0.001). In conclusion, this study suggests heterogenous depending cause keloid, level may response rated diverse therapeutics.
